The study of multiple biomarkers in bioindicator species is a useful tool to evaluate water quality in addition to physicochemical analysis. The aim of this work was to study the toxicity of water samples from two sites with different anthropogenic impacts (R: near a residential area and FP: close to horticultural farms and industrial waste treatment plants) from Las Catonas sub-basin (Reconquista River basin) in the native gastropod Biomphalaria straminea. Some physicochemical parameters and chlorpyrifos concentration were measured in water samples. Snails were exposed in laboratory conditions 48 h to the water samples and neurotoxicity, behavior, lethality and acetylcholinesterase, carboxylesterase, glutathione S-transferase, glutathione reductase and catalase activities were measured. In water from FP, chlorpyrifos was detected and conductivity and pH were higher than in R. Lethality (60%) and a decrease (30%) in acetylcholinesterase were observed in snails exposed to FP indicating that water contamination causes high toxicity in B. straminea.

Lugano Lake is located in an Ecological Reserve of Buenos Aires City. Biomonitoring of its water quality is essential due to its importance as a place for recreation and protection of native species. Biomphalaria straminea is a native hermaphrodite aquatic gastropod that inhabits different freshwater bodies of Argentina and was recently selected as a potential bioindicator. We propose this study as a first approach to assessing specific organisms' use in biomonitoring of urban wild reserves, and the usefulness of reproduction assays. B. straminea survival, behavior, reproduction success and offspring survival after the exposure to water samples from Lugano Lake (L1, L2, and L3) were evaluated. Temperature, pH, conductivity and dissolved oxygen were registered in situ. Samples were transported to the laboratory and chemical analysis and bioassays were performed using 20 snails per site. A control group with tap water was added. Egg masses were separated, exposed individually and observed daily using a stereoscopic microscope. After hatching, juveniles were placed in tap water and offspring survival was registered at the first, second, third and fourth months after the beginning of the assay. High levels of conductivity, turbidity and nutrients were obtained. Ammonium and nitrite were higher than the guideline level for the protection of aquatic life. During the bioassay 20% of the snails (L2 and L3) showed abnormally protruding of the head-food region. The number of eggs and embryonated eggs per mass did not differ between treatments. Egg masses exposed to water samples from the lake presented overlapping and abnormal eggs and arrested embryos. Besides, low % of hatching (L1: 33%, L2: 42%, and L3: 16%) and juvenile survival after the first (L1:14%; L2:78%) and second month (L1: 60%) were noted. In the control group, 85% of hatching and 100%-90% of survival were observed. Our results suggests the presence of pollutant in the lake. B. straminea seems to be a sensitive local species. Biomphalaria spp. reproduction assays can provide a valuable endpoint for toxicity and risk assessments and a usefulness tool for biomonitoring water quality.

Gills represent one of the major sites of gas exchange of fish, consequently they are in continuous close contact with the aquatic environment and its pollutants. In the present study the effects on gills of pejerrey fish, Odontesthes bonariensis, under glyphosate-based herbicide subchronic exposure were analyzed. Adult animals were exposed to sublethal concentrations of a glyphosate-based commercial formulation (1 and 10 PMG mg L-1, PMG: glyphosate active ingredient) for 15 days, while control group was maintained in rearing water. Ultrastructural changes in gills were observed by scanning electron microscopy (SEM). The composition of the surface epithelium and specific surface area were determined by energy dispersive spectroscopy (EDS) and N2 (g) adsorption-desorption isotherms, respectively. The herbicide exposure induced severe alterations in gill ultrastructure, as shown in the SEM micrographs. Accordingly, an increase in surface area of the gills of exposed animals was determined. These results support that gills parameters of freshwater fish are sensitive morphological biomarkers for glyphosate exposure.

The herbicide glyphosate [N- (phosphonomethyl) glycine; PMG] and the insecticide chlorpyrifos [O, O-diethyl O- (3,5,6-trichloro-2-pyridinyl) -phosphorothioate, CPF] are widely used in agricultural practices around the world and can reach aquatic environments. Therefore, it is necessary to characterize the toxicity of these pesticides on non-target species. The use of biomarkers as a tool to assess responses of organisms exposed to pollutants requires the understanding of their natural fluctuation and the dose-response relationship. In the present work, the effect of the exposure to PMG and CPF on the acetylcholinesterase activity (AChE, biomarker of neurotoxicity) in Cnesterodon decemmaculatus, a native teleost, was evaluated in different environmental conditions. Semi-static bioassays of acute toxicity were carried out under controlled conditions during the four weather seasons of the year using animals of homogeneous size. Circannual rhythms in the basal levels of AChE activity in homogenates of the anterior section were confirmed. Statistically significant average inhibition of AChE activity (47.1 ±â€¯0.7% for 1 µg CPF × L-1; 69.7 ±â€¯2.5% for 5 µg CPF × L-1; 23.1 ±â€¯1.1% for 1 mg PMG × L-1 and 32.9 ±â€¯3.3% for 10 mg PMG × L-1) was determined during summer, winter and spring weather seasons. Interestingly, animals exhibit an increased susceptibility to exposure during the autumn season (inhibition of 55.4 ±â€¯0.6% for 1 µg CPF × L-1; 81.9 ±â€¯3.3% for 5 µg CPF × L-1; 41.4 ±â€¯1.7% for 1 mg PMG × L-1 and 61.1 ±â€¯0.3% for 10 mg PMG × L-1). A different sensitivity of the enzyme between seasons was evaluated by in vitro tests. The inhibition pattern for chlorpyrifos-oxon (CPF-oxon, the active metabolite of CPF) was not affected when test was performed using homogenates of unexposed specimens of summer or autumn. Otherwise, PMG in vitro inhibitory effect was not observed in a wide range of concentrations. The results confirm that AChE activity is a sensitive biomarker for exposure to CPF and PMG, even at environmentally relevant concentrations. Finally, this work highlights the existence of seasonal variations in the dose-response relationship, which could be due to variations in the metabolism of the pollutants.

17ß-Estradiol (E2) and synthetic 17α-Ethinylestradiol (EE2) are estrogenic compounds present in surface waters as a consequence of municipal sewage discharges. The aim of this study was to evaluate the effects of E2, EE2 and its mixtures on different reproductive parameters and embryo-larval survival in pejerrey fish (Odontesthes bonariensis). In order to analyze the effect of these compounds on sperm quality, fertilization%, embryo-larval survival (%), and the point of no return (PNR), different assays were performed using concentrations 175, 350, 700 and 1400 ng/L of E2; 22.5, 45, 90 and 180 ng/L of EE2 and mixtures M1 (175 E2+22.5 EE2, ng/L), M2 (350 E2+45 EE2, ng/L), M3 (700 E2+90 EE2, ng/L) and M4 (1400 E2+180 EE2 ng/L). No significant differences in motility parameters were observed between E2 and EE2 treatments and the control group. However, a significant decrease in motility% was recorded for all mixtures tested compared with the control samples. For fertilization%, only sperm activated with M4 showed a significant decrease compared with the control group. In the case of embryo survival, there was only a significant decrease in the highest concentration of EE2 compared with the control group. For the mixtures, M3 is the one that had the most adverse effect on embryo survival. In larval survival, there was a significant decrease in concentration 175 and 700 ng/L of E2 compared with the control group. In EE2 treatments, the ones with a significant reduction in larval survival were concentration 45 and 90 ng/L. And for the mixture treatments, M1, M3 and M4 had a significantly lower larval survival than the control group. In comparison to other treatments, M1 demonstrated a significant difference in PNR when compared with the control group. The results obtained demonstrated that the exposure to mixtures of E2 and EE2 affected fish sperm motility, fertilization% and, embryo and larval survival even at relevant environmental concentrations highlighting the necessity of considering the effects of pollutants mixtures in ecotoxicological studies.

Water pollution by agrochemicals is currently one of the most critical problems for the conservation of aquatic ecosystems. Glyphosate [N-(phosphonomethyl) glycine); PMG] is the main broad-spectrum post emergence herbicide used for the control of a wide range of pests in soybean crops. Adenylate energy charge (AEC) reflects the energy balance of the cells, a measure of the energy available from the adenylate pool: adenosine triphosphate (ATP), adenosine diphosphate (ADP) and adenosine monophosphate (AMP). Background adenylates, phosphagens and the AEC index of two year old Odontesthes bonariensis were determined in some tissues by HPLC, and the impact on subcellular energy balance of sublethal glyphosate-based herbicide exposure was analyzed. The doses used were 0 (control tank), 1 or 10mg PMGL(-1), trials were carried out during 15 days. AEC values in brain, liver and muscle from control fish were 0.37 ± 0.02, 0.49 ± 0.05 and 0.56 ± 0.03, respectively (means ± SEM). While brain ATP concentrations were undetectable (hence low values of AEC), the muscle tissue showed the highest concentrations of the more energetic molecules: 0.18 µmole ATP g(-1) and 8 µmole phosphocreatine g(-1) (PCrg(-1)). In the brain, no significant changes were detected in exposed fish compared to controls. Instead, in both the liver and muscle of animals exposed to the highest concentration of the herbicide, significant changes in the AEC (reduction of 26% and 15%, p<0.05) with respect to the control group were determined. Chronic exposure (15 days) of Odontesthes bonariensis to 1 and 10mgL(-1) of formulated glyphosate did not affect brain AEC. However, the highest concentration of the herbicide produced a significant decrease in liver and muscle AEC manifesting adverse sublethal effects on the energy metabolism. These results suggest the usefulness of AEC as a biomarker of fish glyphosate exposure.

The use of biomarkers as a tool to assess responses of organisms exposed to pollutants in toxicity bioassays, as well as in aquatic environmental risk assessment protocols, requires the understanding of the natural fluctuation of the particular biomarker. The aim of this study was to characterize the intrinsic variations of acetylcholinesterase (AChE) activity in tissues of a native freshwater teleost fish to be used as biomarker in toxicity tests, taking into account both seasonal influence and fish size. Specific AChE activity was measured by the method of Ellman et al. (1961) in homogenates of fish anterior section finding a seasonal variability. The highest activity was observed in summer, decreasing significantly below 40% in winter. The annual AChE activity cycle in the anterior section was fitted to a sinusoidal function with a period of 11.2 months. Moreover, an inverse relationship between enzymatic activity and the animal size was established. The results showed that both the fish length and seasonal variability affect AChE activity. AChE activity in fish posterior section showed a similar trend to that in the anterior section, while seasonal variations of the activity in midsection were observed but differences were not statistically significant. In addition, no relationship between AChE and total tissue protein was established in the anterior and posterior sections suggesting that the circannual rhythms observed are AChE-specific responses. Results highlight the importance of considering both the fish size and season variations to reach valid conclusions when AChE activity is employed as neurotoxicity biomarker.

The toxic effect of sublethal concentrations (1, 17.5 and 35 mg L(-1)) of pure glyphosate was evaluated on acetylcholinesterase (AChE) activity in the fish species, Cnesterodon decemmaculatus. Acute bioassays (96 h) under laboratory conditions were conducted and homogenates for each specimen corresponding to the anterior, middle and posterior body sections were performed. Fish survival was 100%, even at the highest concentration tested (35 mg L(-1)), in accordance with the low lethal toxicity reported for glyphosate. However, a significant inhibitory effect on AChE activity was recorded even for the lowest herbicide concentration tested (1 mg L(-1)), in the homogenates corresponding to the anterior body section. The inhibition ranged from 23 to 36%. The analytical determination of glyphosate in assay media by ion chromatography, was used to verify its stability. These results indicate that AChE-a neurotoxicity biomarker-in C. decemmaculatus may be affected by exposure to environmentally relevant concentrations of glyphosate.

Calcium (Ca(2+)) is an absolute requirement for a decisive sperm function event: the acrosome reaction (AR). Physiologically, sperm capacitation is a prerequisite for this specialized exocytosis and both events are intimately related. In an effort to separate capacitation from AR, we have been using a modified sperm incubation medium where Ca(2+) is replaced by Strontium (Sr(2+)). The aim of this report is to analyze with more detail the difference between sperm incubated with Ca(2+) or Sr(2+) in several events. We found that sperm undergo the capacitation-related changes in the chlortetracycline (CTC) pattern and tyrosine phosphorylation, and also bind to the zona pellucida (ZP) when using Sr(2+)-instead of Ca(2+)-containing media. However, the spontaneous AR typical of hamster sperm does not take place in Sr(2+)-medium, even if sperm are previously capacitated with Ca(2+). Nevertheless, Sr(2+) was able to sustain AR when cells were treated with thapsigargin or depolarized with K(+) in Na(+)-depleted medium. Considering that the absence of Na(+) increased spontaneous AR in Sr(2+)-medium, we tested whether Na(+)-transport systems could be involved in the inability of Sr(2+)-incubated sperm to undergo AR. We found that when sperm incubated in Sr(2+)-medium are treated with amiloride to inhibit epithelial Na(+) channel (ENaC), they are able to undergo spontaneous AR. The same result was obtained when analyzing AR on the ZP. On the contrary, addition of ouabain (a Na(+)/K(+)-ATPase inhibitor) or DIDS (a Na(+)/HCO3(-) co-transporter inhibitor) showed no effect. These results suggest that, differing from what happens in Ca(2+)-incubated sperm, cells incubated in Sr(2+)-modified medium would have an active ENaC.
